    Dusty ERO Search behind Two Massive Clusters

    We performed deep K'-band imaging observations of 2 massive clusters, MS 0451.6-0305 at z = 0.55 and MS 0440.5+0204 at z = 0.19, for searching counterparts of the faint sub-mm sources behind these clusters, which would provide one of the deepest extremely red object(ERO) samples. Comparing our near-infrared images with optical images taken by the Hubble Space Telescope and by the Subaru Telescope, we identified 13 EROs in these fields. The sky distributions of EROs are consistent with the previous results, that there is a sign of strong clustering among detected EROs. Also, the surface density with corrected lensing amplification factors in both clusters are in good agreement with that derived from previous surveys. We found 7 EROs and 3 additional very red objects in a small area (\sim 0.6 arcmin^2) of the MS 0451.6-0305 field around an extended SCUBA source. Many of their optical and near-infrared colors are consistent with dusty star-forming galaxies at high redshifts(z \sim 1.0-4.0), and they may be constituting a cluster of dusty starburst galaxies and/or lensed star-forming galaxies at high redshift. Their red J-K' colors and faint optical magnitudes suggest they are relatively old massive stellar systems with ages(>300 Mega years) suffering from dust obscuration. We also found a surface-density enhancement of EROs around the SCUBA source in the MS 0440.5+0204 field.Comment: 19 pages, 11 figures, Latex(using pasj00.cls).     Ratio of von Willebrand factor propeptide to ADAMTS13 is associated with severity of sepsis.

    Von Willebrand factor (VWF)-cleaving protease (ADAMTS13) cleaves ultralarge VWF (ULVWF) secreted from endothelium and by which is regulating its physiologic function. An imbalance between ULVWF secretion and ADAMTS13 level occurs in sepsis and may cause multiple organ dysfunction. We evaluated the association between the VWF-propeptide (VWF-pp)/ADAMTS13 ratio and disease severity in patients with severe sepsis or septic shock. In 27 patients with severe sepsis or septic shock and platelet count less than 120,000/μL, we measured plasma VWF, VWF-pp, and ADAMTS13 levels on hospital days 1, 3, 5, and 7. The VWF-pp/ADAMTS13 ratio was increased greater than 12-fold in patients with severe sepsis or septic shock on day 1 and remained markedly high on days 3, 5, and 7 compared with normal control subjects. The VWF-pp/ADAMTS13 ratio significantly correlated with Acute Physiology and Chronic Health Evaluation II score on days 1 and 5; Sepsis-related Organ Failure Assessment score on days 1, 3, and 5; maximum Sepsis-related Organ Failure Assessment score and tumor necrosis factor α level on days 1, 3, 5, and 7; and creatinine level on days 1, 5, and 7. Patients with greater than stage 1 acute kidney injury had significantly higher VWF-pp/ADAMTS13 ratio than patients without acute kidney injury. In summary, the VWF-pp/ADAMTS13 ratio was associated with disease severity in patients with severe sepsis or septic shock and may help identify patients at risk for multiple organ dysfunction by detecting severe imbalance between ULVWF secretion and ADAMTS13 level.博士(医学)・乙第1318号・平成25年7月22

    A Terrestrial SER Estimation Methodology based on Simulation coupled with One-Time Neutron Irradiation Testing

    電子機器の信頼性評価の迅速化に光明 --様々な中性子施設で半導体ソフトエラー評価を可能にする技術を開発--. 京都大学プレスリリース. 2023-06-08.Terrestrial soft error rates (SERs) are generally estimated by performing an experiment using spallation neutron beam with the energy spectrum being similar to that of the terrestrial neutrons or at least four measurements using various (quasi-)mono-energetic neutron and/or proton sources to determine the parameters of the Weibull function. We here propose a method to estimate the terrestrial SERs based on simulation coupled with one-time neutron irradiation testing which can be applied to various kinds of neutron sources. In this method, the dependences of single event upset (SEU) cross sections on the neutron energy and the critical charge are calculated by simulation using Particle and Heavy Ion Transport code System (PHITS). The critical charge is used as the only calibration parameter, which is adjusted to reproduce the SER measured by one-time neutron irradiation. The validity of our method is investigated for 65-nm bulk SRAMs with the measured data using various neutron sources in Japan. Our method generally provides the reasonable terrestrial SERs compared with those obtained by the Weibull function method. This result indicates the feasibility of evaluating the terrestrial SER using one of the various neutron sources available all over the world, including those not dedicated to SER measurement. We also investigate the necessity of the elaborated geometry of device under test (DUT) for the accuracy of the simulation. It is shown that detailed material compositions of DUT are not necessary in our method except when the one-time irradiation is performed using the neutron source that contains a high-quantity of low-energy neutrons below 8 MeV. Furthermore, we confirm that the configuration of the sensitive volume can be simplified without sacrificing the estimation accuracy. These simplifications in the simulation help to reduce the modeling and calculation cost in SER estimation

    ABSTRACT We and other investigators obtained evidence that platelets stimulate endothelin-i (ET-i) production at both message and protein levels in vascular endothelial cells (EC5),and that plate let-derived transforming growth factor-f31 (TGF-j3i) is respon sible for this stimulation. In the present study, we examined the effects of acidification or heat treatment, known to activate latent TGF-f31, on the platelet supernatant-induced Er-i pro duction in cultured porcine aortic ECs. Supematant of platelets (6.0 x 1O@ platelets/mI) aggregated by adenosine diphosphate contained large amounts of TGF-pi, but were almost in a latent form, and the proportion of active TGF-j31 in the supematant was increased markedly in the case of acidification or heat treatment. These treatments also significantly potentiated the supematant-induced stimulation of prepro Er-i mRNA expres sion and the Er-i release in ECs. Purified TGF-@i also en hanced Er-i release, dose-dependently, but the enhancement declined at the higher concentrations. Thus, powerful stimula tion of Er-i production by platelet supematant after acidifica tion or heat treatment cannot be explained only by increments in active TGF-131. The supematant-induced stimulation of Er-i synthesis was significantly inhibited by concomitant treatment of TGF-@i neutralizing antibody, but this inhibition was incom plete even at a concentration that abolished TGF-31 -induced maximal stimulation. These resutts suggest that platelet-induced stimulation and subsequent acidification and heat treatment induced potentiation on endothelial Er-i production depend closely on release and activation of TGF-@i derived from plate lets. However, when TGF-fii concentration is increased, this peptide may further stimulate Er-i production, probably through interactions with other platelet-derived substances

    Decomposition of the Superwind in M82

    We present new optical images (BB, VV, and Hα\alpha) of the archetypical starburst/superwind galaxy M82 obtained with the 8.2 m Subaru Telescope to reveal new detailed structures of the superwind-driven nebula and the high-latitude dark lanes. The emission-line nebula is decomposed into (1) a ridge-dominated component comprising numerous filament/loop sub-structures whose overall morphology appears as a pair of narrow cylinders, and (2) a diffuse component extended over much wider opening angle from the nucleus. We suggest that these two components have different origins. The ridge-dominated component appears as a pair of cylinders rather than a pair of cones. Since this morphological property is similar to that of hot plasma probed by soft X-ray, this component seems to surround the hot plasma. On the other hand, the diffuse component may arise from dust grains which scatter stellar light from the galaxy. Since inner region of this component is seen over the prominent ^^ ^^ X"-shaped dark lanes streaming out from the nuclear region and they can be reproduced as a conical distribution of dust grains, there seems to be a dusty cold outflow as well as the hot one probed by soft X-ray and shock-excited optical emission lines. If this is the case, the presence of such high-latitude dust grains implies that neutral gaseous matter is also blown out during the course of the superwind activity.Comment: 12 pages, 6 figures.     A diagnostic marker for superficial urothelial bladder carcinoma : lack of nuclear ATBF1 (ZFHX3) by immunohistochemistry suggests malignant progression

    Background: Pathological stage and grade have limited ability to predict the outcomes of superficial urothelial bladder carcinoma at initial transurethral resection (TUR). AT-motif binding factor 1 (ATBF1) is a tumor suppressive transcription factor that is normally localized to the nucleus but has been detected in the cytoplasm in several cancers. Here, we examined the diagnostic value of the intracellular localization of ATBF1 as a marker for the identification of high risk urothelial bladder carcinoma. Methods: Seven anti-ATBF1 antibodies were generated to cover the entire ATBF1 sequence. Four human influenza hemagglutinin-derived amino acid sequence-tagged expression vectors with truncated ATBF1 cDNA were constructed to map the functional domains of nuclear localization signals (NLSs) with the consensus sequence KR[X10-12]K. A total of 117 samples from initial TUR of human bladder carcinomas were analyzed. None of the patients had received chemotherapy or radiotherapy before pathological evaluation. Results: ATBF1 nuclear localization was regulated synergistically by three NLSs on ATBF1. The cytoplasmic fragments of ATBF1 lacked NLSs. Patients were divided into two groups according to positive nuclear staining of ATBF1, and significant differences in overall survival (P = 0.021) and intravesical recurrence-free survival (P = 0.013) were detected between ATBF1+ (n= 110) and ATBF1− (n=7) cases. Multivariate analysis revealed that ATBF1 staining was an independent prognostic factor for intravesical recurrence-free survival after adjusting for cellular grading and pathological staging (P = 0.008). Conclusions: Cleavage of ATBF1 leads to the cytoplasmic localization of ATBF1 fragments and downregulates nuclear ATBF1. Alterations in the subcellular localization of ATBF1 due to fragmentation of the protein are related to the malignant character of urothelial carcinoma. Pathological evaluation using anti-ATBF1 antibodies enabled the identification of highly malignant cases that had been overlooked at initial TUR.     Bolaamphiphile Analogues of 12-bis-THA Cl2 Are Potent Antimicrobial Therapeutics with Distinct Mechanisms of Action against Bacterial, Mycobacterial, and Fungal Pathogens

    12-Bis-THA Cl2 [12,12'-(dodecane-1,12-diyl)-bis-(9-amino-1,2,3,4-tetrahydroacridinium) chloride] is a cationic bolalipid adapted from dequalinium chloride (DQC), a bactericidal anti-infective indicated for bacterial vaginosis (BV). Here, we used a structure-activity-relationship study to show that the factors that determine effective killing of bacterial, fungal, and mycobacterial pathogens differ, to generate new analogues with a broader spectrum of activity, and to identify synergistic relationships, most notably with aminoglycosides against Acinetobacter baumannii and Pseudomonas aeruginosa, where the bactericidal killing rate was substantially increased. Like DQC, 12-bis-THA Cl2 and its analogues accumulate within bacteria and fungi. More hydrophobic analogues with larger headgroups show reduced potential for DNA binding but increased and broader spectrum antibacterial activity. In contrast, analogues with less bulky headgroups and stronger DNA binding affinity were more active against Candida spp. Shortening the interconnecting chain, from the most lipophilic twelve-carbon chain to six, improved the selectivity index against Mycobacterium tuberculosis in vitro, but only the longer chain analogue was therapeutic in a Galleria mellonella infection model, with the shorter chain analogue exacerbating the infection. In vivo therapy of Escherichia coli ATCC 25922 and epidemic methicillin-resistant Staphylococcus aureus 15 (EMRSA-15) infections in Galleria mellonella was also achieved with longer-chain analogues, as was therapy for an A. baumannii 17978 burn wound infection with a synergistic combination of bolaamphiphile and gentamicin. The present study shows how this class of bolalipids may be adapted further to enable a wider range of potential applications. IMPORTANCE While we face an acute threat from antibiotic resistant bacteria and a lack of new classes of antibiotic, there are many effective antimicrobials which have limited application due to concerns regarding their toxicity and which could be more useful if such risks are reduced or eliminated. We modified a bolalipid antiseptic used in throat lozenges to see if it could be made more effective against some of the highest-priority bacteria and less toxic. We found that structural modifications that rendered the lipid more toxic against human cells made it less toxic in infection models and we could effectively treat caterpillars infected with either Mycobacterium tuberculosis, methicillin resistant Staphylococcus aureus, or Acinetobacter baumannii. The study provides a rationale for further adaptation toward diversifying the range of indications in which this class of antimicrobial may be used
